If you are starting from the San Gimignano's main entrance at Porta San Giovanni, walk straight along Via San Giovanni. You will soon pass a few shops and cafes. Continue walking straight until you reach Piazza della Cisterna, which will be on your left. This is a beautiful square surrounded by medieval towers, and it’s hard to miss.

From Piazza della Cisterna, if you are near the Collegiata di San Gimignano (the main church), walk down Via della Madonna, which is adjacent to the church. Follow this road, and it will lead you directly to Piazza della Cisterna. You will see the square once you reach the end of the street.
